Changes in the concentrations of deoxynucleotides in human lymphocytes treated in vitro and genotoxic agents

By Ferraro, P. - Biasin, M.R. - Celotti, L. - Bianchi, V.

We studied the toxic and genotoxic effects of UV light and an alkylating agent (methyl methanesulphonate; MMS) in human peripheral blood lymphocytes (PBL) in G(o) and after PHA stimulation. We measured DNA repair synthesis and replication in treated PBL together with deoxynucleoside triphosphate pool and ATP concentrations. In addition to its genotoxic action, MMS induced a strong toxic effect in cycling cells, as shown by a marked depletion of nucleotide pools.
